Reference standard or certified reference material?
Two terms used interchangeably that are not the same. What each involves, why the distinction matters on a COA, and which applies to peptide testing.
Both terms describe material used to anchor an analytical measurement, and they are often used interchangeably in supplier copy. They should not be. A certified reference material carries a formal chain of traceability and a stated uncertainty; a reference standard is a well-characterised working material that does the same job in practice without the paperwork. This note explains each, why the distinction matters when reading a COA, and which applies to research peptide testing. All Alphex products are for laboratory and in-vitro research only.

Reference standard (working standard)
A sample of the target compound, of established identity and purity, held by a laboratory and run alongside test samples. Its identity is confirmed by mass spectrometry (often including sequencing by tandem MS), its purity by HPLC under more than one condition, and its content by amino acid analysis or similar. It is stored cold and dark, re-qualified on a schedule, and replaced when it degrades. In peptide HPLC it defines the retention time and peak profile of the target, allows co-injection checks, and if its concentration is known, supports quantification. Certified reference material (CRM)
A reference material produced by a national metrology institute or an accredited reference material producer (ISO 17034), supplied with a certificate stating the certified property value (for example, purity as a mass fraction), its uncertainty, and traceability to the SI. Producing one involves multiple independent methods, inter-laboratory comparison and stability studies. CRMs exist for many pharmaceutical peptides, amino acids and common analytes; they do not exist for most research peptides, because the market is too small to justify the programme.
Why the distinction matters
- Honesty of the COA. A certificate that says "against a certified reference material" for a compound with no CRM in existence is either using the term loosely or misrepresenting. "Against an in-house qualified reference standard" is the accurate description for almost all research peptide testing, and it is a perfectly good one.
- Uncertainty. A CRM gives the measurement a formal uncertainty budget. A working standard gives a comparison whose accuracy rests on how well the standard was qualified. For research use the latter is entirely adequate; for regulated pharmaceutical release it would not be.
- Independence. A laboratory that qualifies its own standard from the same manufacturer's material it is testing has a circularity problem. An independent laboratory with its own standard, sourced or synthesised separately, does not. See third-party versus in-house testing.

Frequently asked questions
Can a laboratory buy a "reference standard" from a peptide vendor?
Yes, and many analytical suppliers sell well-characterised peptide standards. The key is that it is qualified independently of the batches it will be used to test.
Is a pharmacopoeial standard a CRM?
Pharmacopoeial reference standards (USP, EP, BP) are formally established primary standards used for compendial testing; they are closely related to CRMs in rigour and are used where a monograph exists. None exists for the compounds in the Alphex catalogue.
Does the distinction change my results?
