Frozen solution versus refrigerated solution versus dry powder
Stability runs in a clear order. Lyophilised powder in a sealed vial at -20 C is the most stable state by a wide margin: essentially no water is available for hydrolysis and the molecule is immobilised. Frozen solution at -20 C is next: chemical reactions slow enormously, but the peptide has already been through the stress of dissolving and will go through the stress of thawing. Refrigerated solution at 2-8 C is the least stable of the three, with hydrolysis, deamidation and oxidation all proceeding slowly but measurably over weeks. So the rule is: keep powder frozen until you need it, reconstitute what you can use, and freeze the rest as aliquots.
What freezing costs
- Freeze concentration. As ice forms, the remaining liquid becomes more concentrated in peptide, salt and preservative, and its pH can shift. Some peptides aggregate under those conditions.
- Ice-crystal stress. Slow freezing produces large crystals that shear structures; fast freezing produces small ones. For short research peptides this is a minor effect, but for large or lipidated peptides such as GLP-3 (RT) it can promote aggregation.
- Adsorption on thaw. Thawed peptide re-equilibrates with the tube walls; over several cycles a measurable fraction can be lost, particularly at low concentration in ordinary polypropylene.
- Preservative behaviour. Benzyl alcohol in bacteriostatic water tolerates freezing, but the preservative is doing nothing while frozen and resumes on thaw; the 28-day guidance for an opened bacteriostatic vial does not reset because you froze it.
None of these is a reason not to freeze. They are reasons to freeze once.
The aliquot routine
- Reconstitute as described in the step-by-step method. Let the solution stand until fully clear.
- Decide the volume you use per session or per experiment. That is your aliquot size; err on the small side.
- Dispense into labelled low-binding tubes with a calibrated pipette. Cap immediately.
- Freeze at -20 C. A -80 C freezer is fine and slightly better for long storage but not necessary for the compounds in this catalogue.
- Record on the box: peptide, batch number, concentration, diluent, date frozen, number of aliquots.
- To use: thaw one aliquot at room temperature or in the hand, mix by gentle inversion, spin briefly to collect the volume, use, and discard any remainder rather than refreezing.

- GHK-Cu: freezes well; keep dark. Check the colour on thaw; blue is intact.
