Benzyl alcohol in bacteriostatic water, explained

What the 0.9% preservative does and does not do, when it interferes with assays or HPLC, and why the vial is discarded 28 days after opening.

The only thing that separates bacteriostatic water from sterile water is 0.9% benzyl alcohol, and that small addition is responsible for most of the practical questions researchers have about the reagent: what it does, what it does not do, when it interferes with an assay, and why the vial is discarded 28 days after opening. What benzyl alcohol is

Benzyl alcohol (C7H8O, CAS 100-51-6) is an aromatic alcohol: a benzene ring carrying a CH2OH group. It is a colourless liquid, moderately soluble in water (about 4% at room temperature), and has been used as an antimicrobial preservative in multi-dose aqueous preparations for a century. At 0.9% w/v it is well below its solubility limit and stable in solution for years in a sealed vial.

What it does

Benzyl alcohol is bacteriostatic: it inhibits the growth of bacteria introduced during repeated sampling by disrupting membrane function and protein synthesis. In a vial punctured many times over several weeks, that inhibition is the difference between a usable stock and a cloudy one. Its activity is broad against common bacteria and modest against fungi and spores. It is not sporicidal.

What it does not do

  • It does not sterilise a solution that is already contaminated. Preservatives suppress growth; they are not a substitute for aseptic technique.
  • It does not remove or neutralise endotoxin, which is a component of dead bacterial cell walls, not a living organism.
  • It does not stabilise the peptide chemically. Hydrolysis, deamidation and oxidation proceed at the same rate with or without it. Its contribution to shelf life is entirely microbiological.
  • It does not act while frozen, and its 28-day clock does not reset because the vial was frozen.

When it interferes

  • Cell culture. At 0.9% benzyl alcohol is cytotoxic to most cell lines. A peptide stock made in bacteriostatic water and diluted 1:1000 into medium delivers 0.0009%, which is generally tolerated, but calculate the final concentration for your dilution factor and include a vehicle control at the same level.
  • UV detection. Benzyl alcohol absorbs strongly at 220 nm and around 257 nm. On reverse-phase HPLC it elutes as a sharp, well-retained peak. If you are analysing your own stocks, prepare the analytical sample in water or mobile phase, or identify and exclude the benzyl alcohol peak from the purity calculation.
  • Enzyme and receptor assays sensitive to alcohols. Rare at the concentrations reached after dilution, but a vehicle control settles it.
  • Plastics. Benzyl alcohol can partition into some rubbers and soft plastics over long contact, which is one reason for the 28-day rule and for storing in glass or in polypropylene rather than in soft PVC.

The 28-day rule

Once a multi-dose vial has been punctured, the convention is to discard it 28 days later. The reasoning: each entry risks introducing organisms; the preservative's margin is finite; and benzyl alcohol slowly migrates into the stopper, lowering its concentration in the water. Frequently asked questions

Can I add benzyl alcohol to my own buffer?

Yes, to 0.9% w/v, using a laboratory-grade reagent, if you want a preserved buffer for multi-use peptide stocks. Check compatibility with the assay as above.

Is 0.9% a specific standard?

It is the long-established concentration for bacteriostatic water and saline; higher adds toxicity without useful gain, lower loses efficacy.

Does it affect peptide solubility?

Marginally, if at all, at 0.9%. It is not a co-solvent in the sense that DMSO or ethanol are.
